순위 조정 안내 — 반응성 구조 412건 (시즌 1~4 전체)

전 시즌 순위표에 구조 경보 축을 추가하고, 약이 될 수 없는 반응성 구조를 순위에서 제외했습니다. 점수는 그대로 두었고, 항목도 표에 남아 있습니다. 바뀐 것은 등수뿐입니다.

왜 필요했는가

도킹 점수는 잘 달라붙는 분자에 높게 나옵니다. 아무 단백질에나 공유결합으로 달라붙는 친전자체는 그래서 만점에 가까운 결합 점수를 받습니다. 선택성이 아니라 무차별 반응성으로 얻은 점수입니다.

측정해 보니 상위권에 몰려 있었습니다.

시즌 1은 상위 10위가 전부 아지리딘(3원 고리 질소)이었습니다. 전체 모집단 대비 12배 농축이며, 우연으로 설명되지 않습니다. 시즌 3은 α-할로 헤테로원자와 텔루륨이 주를 이뤘습니다.

무엇을 제외했는가

제외 — 아지리딘 · 에폭사이드 · 다이아지린 · 아실할라이드 · 이소시아네이트 · 질소머스타드 · α-할로 헤테로원자 · 이할로메틸 헤테로원자, 그리고 의약품 전례가 없고 도킹 힘장 파라미터도 신뢰할 수 없는 원소(텔루륨 등).

제외하지 않음 — 마이클 수용체 · 셀레늄 · 비소. 이브루티닙·아파티닙·오시머티닙은 마이클 수용체를 가진 승인약이고, 삼산화비소는 급성전골수구성백혈병 승인약입니다. 이들은 표시만 하고 순위를 유지합니다.

시즌 4는 접수 단계에서 공유결합 규칙이 이미 작동하고 있어 해당 항목이 없었습니다.

함께 반영된 것

총점 옆에 ± 를 붙였습니다. 도킹은 실행마다 흔들리고, 저희는 그 흔들림을 재고 있으면서도 순위표에는 평균 하나만 올렸습니다. 이제 저장된 반복 측정값을 200회 재표본하여 95% 구간을 함께 표시합니다.

같은 시즌 안에서도 분자마다 흔들림이 10배 넘게 차이납니다. 평균만 보면 그 차이가 보이지 않습니다. 상위권 일부는 구간이 서로 겹치며, 그 경우 순위는 데이터가 아니라 반올림이 정한 것입니다. 판단에 참고하시기 바랍니다.

앞으로

이 검사는 이제 채점 시점에 적용됩니다. 새 제출은 접수·채점되지만, 해당 구조는 등수를 받지 않고 사유가 표시됩니다.

Ranking adjustment — 412 reactive structures (Seasons 1–4)

We have added a structural alert axis to every season's leaderboard and removed non-drug reactive structures from the rankings. Scores are unchanged and every entry remains on the table. Only the rank changed.

Why

Docking rewards molecules that bind well. An electrophile that binds almost any protein covalently therefore earns a near-maximal binding score — earned by indiscriminate reactivity, not selectivity.

Measured, it was concentrated at the top:

Season 1's entire top 10 carried an aziridine — a 12-fold enrichment over the population, which chance does not explain. Season 3 was dominated by α-halo heteroatoms and tellurium.

What was excluded

Excluded — aziridine, epoxide, diazirine, acyl halide, isocyanate, nitrogen mustard, α-halo heteroatom, dihalomethyl heteroatom, and elements with no medicinal precedent and no trustworthy docking parameters (tellurium and similar).

Not excluded — Michael acceptors, selenium, arsenic. Ibrutinib, afatinib and osimertinib are approved Michael-acceptor drugs; arsenic trioxide is approved for APL. These are flagged and keep their rank.

Scale

Season 1: 270 · Season 2: 26 · Season 3: 116 · Season 4: 0 — 412 total. Season 4 had none because its covalent rule was already active at submission.

Also shipped

A ± now sits beside every total. Docking varies run to run; we were measuring that variation and publishing only the mean. Totals now carry a bootstrap standard deviation and 95% interval over 200 resamples of the stored repeat measurements.

Within a single season the variation differs more than tenfold between molecules — invisible in the mean alone. Some top intervals overlap, and where they do, the ordering is decided by rounding rather than by data. Read the rankings accordingly.

Going forward

The check now runs at scoring time. New entries are accepted and scored, but such structures receive no rank and carry the reason.
